新西兰进境猕猴桃上狭截盘多毛孢菌的检测鉴定

摘要:

在一批来自新西兰猕猴桃果实上的变褐腐烂部位,分离到1株拟盘多毛孢样真菌(pestalotioid fungi)菌株Ta-1726。该病菌的分生孢子纺锤形、直到微弯、有3个隔膜,大小为:16.89~23.70μm×4.88~8.08μm。顶端和基部细胞半透明,中间2个细胞黑棕色。基部无附属物,顶端附属丝单根,或2~5分支。分别采用真菌ITS、EF-1α、β-tubulin和actin基因区域的通用引物进行PCR扩增和测序,测序结果在NCBI上进行BLAST检索,结果显示菌株Ta-1726的4段基因的核苷酸序列与狭截盘多毛孢菌(Truncatella angustata)的同源性达到98%~100%。致病性测定结果表明,菌株Ta-1726可以侵染猕猴桃和苹果。基于上述实验结果,将分离获得的菌株Ta-1726鉴定为T. angustata,这是我国口岸首次在进境的猕猴桃果实上截获该危险性有害生物。

Abstract:

An isolate Ta-1726 similar to pestalotioid fungi was isolated from the brown rotting sites of kiwifruit imported from New Zealand. Morphological descriptions of Ta-1726 revealed that the conidia fusiform,straight to slightly curved 16.89-23.70 ×4.88-8.08 μm,with 3 transverse septa,apical and basal cells subhyaline,median cells dark-brown;basal appendages absent,apical appendages variable and often branched,single or 2-5. The Ta-1726 genes were amplificated and sequenced by universal primers of the internal transcribed spacer(ITS),beta tubulin (β-tubulin),translation elongation factor 1-alpha(EF-1α) and actin gene(ACT). Compared with the identity in the nucleotide sequence of 4 genes of Truncatella angustata from NCBI GenBank databases by blast method,the results showed that the homology were 98%-100%. Pathogenicity tests showed that the kiwifruit and apple could be infected by Ta-1726. Based on morphological characteristics,molecular and pathogenicity test results,the isolate Ta-1726 was identified as T. angustata. This dangerous pest was intercepted on imported kiwifruit for the first time in China.
