糙果苋快速分子检测技术研究

摘要:

糙果苋(Amaranthus tuberculatus)是当前美国农田的一种恶性杂草,已对多种除草剂产生了抗药性,可造成农作物大量减产。进口美国大豆、玉米等粮谷中经常截获糙果苋种子。为了准确、快速地鉴定糙果苋种子,本文设计了糙果苋特异性引物探针,利用核酸快释技术,改进了实时荧光PCR仪的升降温速度,可以在30 min左右准确检测出进境粮谷中携带的单粒糙果苋种子。该技术大大缩短了进口粮谷中糙果苋的检疫鉴定时间,还可应用于口岸一线杂草监测工作。

Abstract:

Amaranthus tuberculatus is a notorious weed in U.S. farmland that has developed resistance to multiple herbicides and can cause significant crop yield losses. Seeds of Amaranthus tuberculatus are frequently intercepted in imported grains such as soybeans and corn from the United States. To enable accurate and rapid identification of Amaranthus tuberculatus seeds,this study designed specific primers and probes,utilized rapid nucleic acid release technology,and optimized the heating-cooling rate of real-time fluorescent PCR instrument. This method can accurately detect a single Amaranthus tuberculatus seed in imported grains within approximately 30 minutes. The technique significantly reduces the quarantine identification time for Amaranthus tuberculatus in imported grains and can also be applied to frontline weed monitoring at ports.
