烟草环斑病毒是我国检疫性有害生物,为防止该病毒传入国内,需建立快速的诊断和检测方法。将烟草环斑病毒的多克隆抗体吸附到磁性纳米颗粒上,研制出磁免疫层析检测试纸条。该试纸条可特异性检测烟草环斑病毒,检测部分提纯病毒的灵敏度为1μg/mL,检测烟草病汁液可稀释1000倍(重量/体积)。该试纸条和线虫传多面体病毒属的番茄环斑病毒、番茄黑环病毒、南芥菜花叶病毒和葡萄扇叶病毒未出现交叉反应,可用于烟草环斑病毒的现场检测。
Abstract:Tobacco ringspot virus(TRSV)is a quarantine pest for China. In order to prevent the introduction of the virus into China, we need to establish a rapidly diagonstic method for use at the point-of-care or in the fi eld. Magnetic immunochromatography assay was developed by coupling magnetic nanoparticles with polyclonal antibody against TRSV. The sensitivity of the method was 1μg/mL for detecting partial purifi ed TRSV. Extract from TRSV infected Nicotiana tabacum cv. White Burley leaf was detected to a dilution of 10-3(W/V). The immunostrip was specifi c to TRSV. There was no cross reaction with other nepoviruses such as Tomato ringspot virus, Tomato black ring virus, Arabis mosaic virus and Grapevine fanleaf virus. The immunostrip can be used for the on-site detection of TRSV.
