为评估ycf1序列对濒危星球属植物的鉴定能力,本研究选取matK和ycf1引物对18份星球属和13份属外近缘物种进行序列扩增及产物测序,同时比较这2对引物扩增效率、测序成功率和序列差异位点。应用DNASTAR Lasergene软件对测得的序列进行校对拼接;使用MEGA-X软件进行序列比对,并基于邻接法构建系统聚类树。结果表明,相较于matK序列,ycf1序列具有更多的变异位点和更优的聚类效果,ycf1序列可作为星球属植物分子鉴定的有效序列片段。
Abstract:In order to evaluate the identification ability of ycf1 sequence for endangered Astrophytum plants,eighteen Astrophytum and thirteen extragenus closely related species were studied,and the primers of matK and ycf1 were amplified and sequenced,at the same time,the amplification efficiency,sequencing success rate,and sequence differences of the two primers were compared. DNASTAR Lasergene software was used to proofread and splice the measured sequences;MEGA-X software was used to sequence alignment and construct the systematic clustering tree based on the adjacency method. Compared to the matK sequence,the ycf1 sequence has more variation sites and better clustering effect,ycf1 sequence can be used as an effective sequence fragment for molecular identification of Astrophytum plants.
